Smyth line (SL) chickens, which develop a depigmenting disorder similar to human vitiligo, produce circulating anti-melanocyte antibodies (Austin, L.M. et al., (1992) The detection of melanocyte autoantibodies in the Smyth chicken model for vitiligo. Clin. Immunol. Immunopathol., 64:112–120). In order to characterize these autoantibodies, we studied the reactivity of cultured chicken, mouse, and human melanocytes, as well as frozen sections of chicken feather follicles and embryonic eyes, against SL serum, employing indirect immunofluorescence. Light Brown Leghorn (LBL) serum was used as a negative control. Chicken (SL and LBL), mouse, and human melanocytes exhibited greater fluorescence with SL serum than with LBL serum (up to a 1:60,000 dilution). The fluorescent pattern was predominant along the perimeter of the cells, suggesting plasma membrane staining. Fluorescence-activated flow cytometry analysis and immunocytochemical localization at the ultrastructural level using intact chicken cells supported this hypothesis. Melanocytes were readily stained in cryosections of regenerating feather follicles and embryonic eyes incubated with SL, but not LBL, serum. In addition, amelanotic melanocytes in albino chicken feathers reacted with SL serum. SL serum also preferentially stained cells emigrating from cultured avian neural tubes and within the dermis of the proliferative germ of regenerating feather follicles suggesting that melanoblasts express the antigens. We conclude that Smyth line serum contains melanocyte autoantibodies that cross-react with mouse and human melanocytes, are able to bind to pigment cells within tissues, and recognize antigens expressed in the cytoplasm and on the surface of melanocytes and melanoblasts.  相似文献

Alterations in global and regional precipitation patterns are expected to affect plant and ecosystem productivity, especially in water‐limited ecosystems. This study examined the effects of natural and supplemental (25% increase) seasonal precipitation on a sotol grassland ecosystem in Big Bend National Park in the Chihuahuan Desert. Physiological responses – leaf photosynthesis at saturating light (Asat), stomatal conductance (gs), and leaf nitrogen [N] – of two species differing in their life form and physiological strategies (Dasylirion leiophyllum, a C3 shrub; Bouteloua curtipendula, a C4 grass) were measured over 3 years (2004–2006) that differed greatly in their annual and seasonal precipitation patterns (2004: wet, 2005: average, 2006: dry). Precipitation inputs are likely to affect leaf‐level physiology through the direct effects of altered soil water and soil nitrogen. Thus, the effects of precipitation, watering treatment, soil moisture, and nitrogen were quantified via multivariate hierarchical Bayesian models that explicitly linked the leaf and soil responses. The two species differed in their physiological responses to precipitation and were differentially controlled by soil water vs. soil nitrogen. In the relatively deeply rooted C3 shrub, D. leiophyllum, Asat was highest in moist periods and was primarily regulated by deep (16–30 cm) soil water. In the shallow‐rooted C4 grass, B. curtipendula, Asat was only coupled to leaf [N], both of which increased in dry periods when soil [N] was highest. Supplemental watering during the wet year generally decreased Asat and leaf [N] in D. leiophyllum, perhaps due to nutrient limitation, and physiological responses in this species were influenced by the cumulative effects of 5 years of supplemental watering. Both species are common in this ecosystem and responded strongly, yet differently, to soil moisture and nitrogen, suggesting that changes in the timing and magnitude of precipitation may have consequences for plant carbon gain, with the potential to alter community composition.  相似文献

Non‐native invasive plants are a widely acknowledged threat to global biodiversity. However, our understanding of the mechanisms underlying plant invasion, and the relative importance of multiple rather than single drivers, remains poor. Here, we provide a case study using time‐series data to reconstruct patterns of change, and field experiments to test for causality. We show how, over a 50‐year period, a series of unrelated human‐induced changes created highly favorable conditions for the non‐native tree mallow (Lavatera arborea) to turn invasive, causing loss of native vegetation and seabird breeding habitat. The combination of three drivers: human‐introduced disease, climate warming and a fisheries‐mediated increase in seabird populations, removed major constraints on plant population growth, (i.e. grazer control, climatic control, germination opportunity and nutrient limitation). Collectively, these changes created optimal conditions for the rapid expansion of tree mallow. The resulting dramatic impact on both the native vegetation and breeding seabirds, notably puffins (Fratercula arctica), exemplifies how non‐native invasive plant species can transform terrestrial ecosystems. While climate change is regarded as a key factor behind plant invasion, we highlight that multiple rather than single factors may be critical to biodiversity loss.  相似文献

High-alpine life forms and ecosystems exist at the limits of habitable environments, and thus, are especially sensitive to environmental change. Here we report a recent increase in the elevational limit of anurans following glacial retreat in the tropical Peruvian Andes. Three species have colonized ponds in recently deglaciated terrain at new record elevations for amphibians worldwide (5244–5400 m). Two of these species were also found to be infected with Batrachochytrium dendrobatidis ( Bd ), an emerging fungal pathogen causally associated with global amphibian declines, including the disappearance of several Latin American species. The presence of this pathogen was associated with elevated mortality rates of at least one species. These results represent the first evidence of upward expansion of anurans to newly available habitat brought about by recent deglaciation. Furthermore, the large increase in the upper limit of known Bd infections, previously reported as 4112 m in Ecuador, to 5348 m in this study, also expands the spatial domain of potential Bd pathogenicity to encompass virtually all high elevation anuran habitats in the tropical Andes.  相似文献

1. This paper reviews and compares the effects of forest fire and timber harvest on mammalian abundance and diversity, throughout successional time in the boreal forest of North America. 2. Temporal trends in mammal abundance and diversity are generally similar for both harvested and burned stands, with some differences occurring in the initiation stage (0–10 years post disturbance). 3. Small mammals and ungulates are most abundant immediately post disturbance, and decrease as stands age. Lynxes and hares utilize mid-successional stands, but are rare in young and old stands. Bats, arboreal sciurids and mustelids increase in abundance with stand age, and are most abundant in old growth. 4. Substantial gaps in the data exist for carnivores; the response of these species to fire and harvest requires research, as predator–prey interactions can affect mammal community structure in both early and late successional stages. 5. The lack of explicit treatment of in-stand forest structure post disturbance, in the reviewed literature made comparisons difficult. Where forest structure was considered, the presence of downed woody material, live residual trees and standing dead wood were shown to facilitate convergence of mammal communities to a pre-disturbance state for both disturbance types. 6. Mammalian assemblages differed considerably between successional stages, emphasizing the importance of maintaining stands of each successional stage on the landscape when implementing forest management strategies.  相似文献

Comparisons between plant species or cultivars differing in root hair length have indicated a major impact of root hairs on the mycorrhizal dependency of plants with respect to phosphate (P) uptake. The current study aimed to investigate this relationship by comparing directly the mycorrhizal dependency of a spontaneous root hairless mutant, brb , in Hordeum vulgare cv Pallas and its wild type. Both brb and wild type were grown at different soil P levels in association with different mycorrhizal fungi. P uptake of brb and wild type was similar at high P levels, but P uptake by non-mycorrhizal brb plants at low P levels was substantially lower than that of the non-mycorrhizal wild-type plants. However, P uptake of the mutant was much increased by mycorrhizas and with one fungus, the additional P uptake was effectively translated into increased plant growth. Roots of the mutant contained typical colonization structures and a radioactive tracer confirmed P transport by the extraradical mycelium. This is the first direct evaluation of the relative effectiveness of root hairs and mycorrhizas. Mycorrhizas effectively substituted root hairs in P uptake, whereas the additional P was most often used less effectively in promoting plant growth than P provided by root hairs.  相似文献

Abstract: Estimating survival of the offspring of marked female ungulates has proven difficult in free-ranging populations yet could improve our understanding of factors that limit populations. We evaluated the feasibility and efficiency of capturing large samples (i.e., >80/yr) of neonate mule deer (Odocoileus hemionus) exclusively from free-ranging, marked adult females using vaginal implant transmitters (VITs, n = 154) and repeated locations of radiocollared females without VITs. We also evaluated the effectiveness of VITs, when used in conjunction with in utero fetal counts, for obtaining direct estimates of fetal survival. During 2003 and 2004, after we placed VIT batteries on a 12-hour duty cycle to lower electronic failure rates, the proportion that shed ≤ 3 days prepartum or during parturition was 0.623 (SE = 0.0456), and the proportion of VITs shed only during parturition was 0.447 (SE = 0.0468). Our neonate capture success rate was 0.880 (SE = 0.0359) from females with VITs shed ≤ 3 days prepartum or during parturition and 0.307 (SE = 0.0235) from radiocollared females without VITs or whose implant failed to function properly. Using a combination of techniques, we captured 275 neonates and found 21 stillborns during 2002-2004. We accounted for all fetuses at birth (i.e., live or stillborn) from 78 of the 147 females (0.531, SE = 0.0413) having winter fetal counts, and this rate was heavily dependent on VIT retention success. Deer that shed VITs prepartum were larger than deer that retained VITs to parturition, indicating a need to develop variable-sized VITs that may be fitted individually to deer in the field. We demonstrated that direct estimates of fetal and neonatal survival may be obtained from previously marked female mule deer in free-ranging populations, thus expanding opportunities for conducting field experiments. Survival estimates using VITs lacked bias that is typically associated with other neonate capture techniques. However, current vaginal implant failure rates and overall expense limit broad applicability of the technique.  相似文献
